Overview

Tirzepatide is a 39-residue acylated peptide built on a GIP scaffold that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide receptor (GIPR) and the glucagon-like peptide-1 receptor (GLP-1R), and has been characterised in the literature as an imbalanced and biased agonist with greater potency at GIPR than at GLP-1R. It is used in vitro to dissect GIPR/GLP-1R co-agonism through cAMP accumulation and β-arrestin recruitment in recombinant cell lines, and has been investigated in preclinical rodent models of energy balance and glucose handling.
