Overview
Semaglutide is an acylated 31-residue GLP-1 analogue and selective GLP-1 receptor agonist; the Aib substitution at position 2 confers resistance to DPP-4 cleavage and the C18 diacid-γGlu-(AEEA)2 side chain mediates reversible albumin binding. In vitro it is used as a reference GLP-1R agonist for receptor binding, cAMP accumulation and β-arrestin recruitment assays, and in rodent studies it has been investigated in preclinical models of energy balance to map hypothalamic and hindbrain circuit engagement.
